Intracellular calcium release at fertilization in the sea urchin egg.

چکیده
Fertilization or ionophore activation of Lytechinus pictus eggs can be monitored after injection with the Ca-sensitive photoprotein aequorin to estimate calcium release during activation. We estimate the peak calcium transient to reach concentrations of 2.5-4.5 & free calcium 45-60 set after activation and to last 2-3 min, assuming equal Ca2+ release throughout the cytoplasm. Calcium is released from an intracellular store, since similar responses are obtained during fertilization at a wide range of external calcium concentrations or in zerocalcium seawater in ionophore activations. In another effort to estimate free calcium at fertilization, we isolated egg cortices, added back calcium quantitatively, and fixed for observation with a scanning electron microscope. In this way, we determined that the threshold for discharge of the cortical granules is between 9 and 16 @f CaZ+. Therefore, the threshold for the in vitro cortical reaction is about five times the amount of free calcium, assuming equal distribution in the egg. This result suggests that transient calcium release is confined to the inner subsurface of the egg.

The Part Played By Inositol Trisphosphate and Calcium in the Propagation of the Fertilization Wave in Sea Urchin Eggs
Sea urchin egg activation at fertilization is progressive, beginning at the point of sperm entry and moving across the egg with a velocity of 5 ~tm/s. This activation wave (Kacser, H., 1955, J. Exp. Biol., 32:451-467) has been suggested to be the result of a progressive release of calcium from a store within the egg cytoplasm (Jaffe, L. E, 1983, Dev. Biol., 99:265-276). The progressive release ...

متن کاملMechanism for Increase in Intracellular Concentration of Free Calcium in Fertilized Sea Urchin Egg
Intracellular free calcium concentration in the sea urchin egg was calculated to increase from 0.1 mM in an unfertilized egg to 1 mM in a fertilized egg 10 min after fertilization, based on measurement of the dissociation constant between free calcium and sea urchin egg homogenate. The dissociation constant between free calcium (dialyzable calcium) and homogenate of sea urchin eggs was measured...
متن کاملMultiple stores of calcium are released in the sea urchin egg during fertilization.
Fertilization initiates a transient increase in intracellular Ca2+ principally by Ca2+ release from intracellular stores. Possible multiple Ca2+ stores and multiple receptor regulation of the same store have been reported. Here we report the presence of at least two independent intracellular Ca2+ stores in the sea urchin egg, which are released during fertilization. Ca2+ release from one store ...
متن کاملCalcium in sea urchin egg during fertilization.
Calcium plays a strikingly important role in two of the major events in developmental biology: cell activation and differentiation. In this review we begin with the location and quantity of intracellular calcium in sea urchin oocytes, and then discuss the changes that occur during fertilization and egg activation, placing special emphasis on the mobilization and redistribution of intracellular ...
